Fx Crown screws on top of receiver

On the dovetail receiver the front most two screws hold barrel in place. What does the rearmost one do ? Just in front of magazine slot.

Hey Mike, it is the hole through which the transfer port hole is drilled.
The set screw is glued in, and the only thing it does is keep the hole closed.

When I shoot my Crown a puff of air comes out of this area, and set screw is free to turn ? Should I put some blue lock tite and screw down snug ?

Yep, it needs to stay put
When it is loose, apply some blue Loctite (243) and screw it back in in such a way it is flush with the top of the receiver.
Don't snug it up, because then you can't take out your barrel anymore
